Growth Promotion Test

To lay down a procedure to check the growth promotion properties of the procured media & also to describes the procedure to be followed to evidence each lot of prepared or autoclaved media support the growth of microorganisms when all the environmental conditions for growth is provided.

Description

1.0    OBJECTIVE:

To lay down a procedure to check the growth promotion properties of the procured media & also to describes the procedure to be followed to evidence each lot of prepared or autoclaved media support the growth of microorganisms when all the environmental conditions for growth is provided.

2.0    SCOPE:

The SOP is applicable to the media used for microbiological analysis in microbiology laboratory.

3.0    RESPONSIBILITY: 

Microbiologist  : To perform the procedure as per SOP.

Head – Quality Control : To ensure the compliance of the SOP.

4.0    PROCEDURE: 

4.1    Liquid broth medium (Growth Promotion / Positive Control): 

4.1.1    Prepare culture dilution as per Growth Promotion Test

4.1.2    Inoculate 1 ml of culture to the sterile culture media and incubate at recommended temperature specified for the organism as per Table -1.

4.1.3    The media complies the growth promotion test if luxuriant growth (turbidity) is observed in the inoculated tubes of liquid media or distinct colony formation on the agar media within 48 hours of  incubation.

4.1.4    If the observations are without any microbial growth discard the batch of medium and reject the entire lot of media..

4.1.5    Observe inoculated media & enter 

4.3    Acceptance criteria :

4.3.1    Micro-organism must show turbidity in the inoculated broth media and distinct colony formation on the solid agar media within 48 hours duration.
